Antibacterial activity of Avocado pear leaf methanolic extract against resistant Salmonella Typhi strains

Abstract
Typhoid fever, caused by Salmonella enterica serovar Typhi, remains a major public health challenge, particularly in developing countries where sanitation is inadequate. The increasing prevalence of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains has rendered conventional antibiotics less effective, necessitating the search for alternative therapies. This study investigated the antibacterial activity of the methanolic leaf extract of Persea americana (avocado pear) against resistant Salmonella Typhi strains. Avocado leaves were collected, shade-dried, pulverized, and extracted with methanol. The antibacterial activity was assessed using the agar well diffusion method, and the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined. Phytochemical screening revealed the presence of bioactive compounds such as flavonoids, phenolics, tannins, alkaloids, and saponins. The results showed that the methanolic extract exhibited dose-dependent antibacterial activity, with inhibition zones ranging from 10–14 mm at higher concentrations (100–200 mg/ml). All test isolates, which had shown resistance to commonly used antibiotics such as amoxicillin, septrin, chloramphenicol and augmentin, were susceptible to the avocado leaf extract at sufficient concentrations. These findings support the ethnomedicinal use of avocado leaves and highlight their potential as a source of novel plant-based antimicrobials for managing resistant Salmonella Typhi infections. Further studies, including bioactive compound isolation and in vivo assays, are recommended to validate their therapeutic potential....
